580 Biodegradable bioadhesive nanoparticle delivery of chemotherapy for the treatment of cutaneous malignancies

Encapsulation of chemotherapeutic agents within biodegradable nanoparticles (NP) allows for gradual drug release from the polylactic acid core. Bioadhesive, degradable NPs can be retained at site administration, maximizing local effects while minimizing systemic side effects. Our bioadhesive NP (BNP) loaded with chemotherapy have previously been proven efficacious in treatment murine PDVC57 squamous cell carcinoma (SCC) tumors, increased intratumoral (i.t.) retention active agent. SBI-111 is a potent anti-tumor agent but considerable toxicity. We examined efficacy i.t. injection BNP encapsulated (BNP-111) pre-clinical melanoma and SCC. determined vitro YUMMER 1.7, an immunogenic line (IC50=0.108±0.05μM) (IC50= 0.055±0.3μM). then assessed therapeutic effect BNP-111 vivo YUMMER1.7 tumors established C57Bl/6 mice. They received single, low dose or vehicle were sacrificed 14 days after treatment. treated had significantly reduced tumor volume (vehicle: 589.3mm3 ±391.2, BNP-111: 108.6 mm3 ±151.7, p=0.0095).
